Research project
Unanticipated drug-drug interactions (DDI) at the level of phase I-enzymes, in particular cytochrome P450’s, are a major cause of adverse drug reactions in human. The predictive value of the non-cellular in vitro models which are used to predict DDI at the level of cytochrome P450s for some drugs is limited due to the absence of phase II enzymes, leading to strong under- or over-prediction of DDIs observed in vivo. The aim of the project is to develop novel, enhanced in vitro ADME models to explore the interplay between phase I and phase II enzymes in more detail.

Tasks
Main Tasks:
•  Heterologous (co)expression of human phase I (P450’s) and phase II (UGT, SULT, GST) enzymes; in bacterial and mammalian cell systems;
•  metabolite profiling by HPLC/LC-MS/MS of incubations of drugs with (mixtures of) recombinant human phase I and phase II enzymes;
•  profiling of inhibitory properties of phase I and corresponding phase II metabolites towards the major drug metabolizing P450s using novel on-line affinity screening methodologies;
•  evaluation of inhibitory properties of drug metabolites in cellular context.

Requirements
The highly motivated candidate must hold an MSc degree in Pharmaceutical Sciences, Biomolecular Sciences, Biomedical Sciences, Chemistry or a related field. The candidate should have less than 4 years of research experience and must not have resided or carried out their main activity in the Netherlands for more than 12 months in the 3 years immediately prior to their recruitment.

This project is part of the EU-funded project ARIADME: Analytical Research in ADME profiling (http://pharm.kuleuven.be/pharbio/ariadme). ARIADME is a Marie Curie project which involves 4 academic centers and 10 industrial partners. The PhD candidate will be enrolled in the PhD-program of AIMMS (aimms.vu.nl) and is expected to contribute to network wide training events of the ARIADME project.

The appointment will be initially for one year. After satisfactory evaluation of the initial appointment, it can be extended for a total duration of four years.
